摘    要:细胞间的变异或异质性是干细胞群的基本和内在特征。传统的组学分析是在混合细胞群体上进行,导致这些差异被掩盖。近年来,单细胞基因组、表观基因组、转录组测序等技术发展迅速,为全面解析细胞异质性和识别不同表型细胞类型提供了强有利工具。这些方法应用于不同类型的干细胞,包括多能干细胞和组织特异性干细胞,在干细胞领域取得了令人兴奋的新发现。本文综述了单细胞组学测序技术的最新进展,并对单细胞组学测序的方法和应用进行了展望。

Abstract:Cell-to-cell variation and heterogeneity are fundamental and intrinsic characteristics of stem cell populations, but these differences are masked when bulk cells are used for traditional omic analysis. These technologies, including single-cell genome, epigenome, and transcriptome sequencing technologies, have been developing rapidly in recent years. Single-cell sequencing technologies serve as powerful tools to dissect cellular heterogeneity comprehensively and to identify distinct phenotypic cell types. The application of these methods to different types of stem cells, including pluripotent stem cells and tissue-specific stem cells, has led to exciting new findings in the stem cell field. In this review, we discuss the recent progress as well as future perspectives in the methodologies and applications of single-cell omic sequencing technologies.
