反应停对人癌裸鼠肿瘤及新血管形成的影响

目的:观察反应停对人卵巢癌细胞(OVCAR-3)及人结肠癌细胞(HCT-8)生长及肿瘤组织内新血管形成的影响。方法:建立人癌裸鼠异种移植的模型,将接种肿瘤后的裸鼠分别灌胃给予不同剂量的反应停,并与正常对照组进行比较,观察肿瘤生长的情况;采用免疫组化检验方法检测肿瘤组织中微血管密度(MVD)。结果:对人癌裸鼠OVCAR-3肿瘤,反应停中、高剂量组均可显著抑制肿瘤生长(P<0.05、P<0.01),对HCT-8的生长却无明显影响,但反应停均呈剂量依赖性降低上述肿瘤组织MVD。结论:反应停的抗肿瘤作用可能与抗新血管形成作用无关。

Effect of Thalidomide on Angiogenesis in Implanted Human Tumors of Nude Mice

OBJECTIVE:To observe the effects of thalidomide in the growth of the human ovarian cancer cells (OVCAR-3) and HCT-8 as well as the angiogenesis in tumor tissue through building a model of heterograft of nude mice with human tumors. METHODS: After implanting human tumors in nude mice for modeling, the inoculated nude mice were intragastrically administered different dose of thalidomide, with tumor growth condition observed and compared with control group. The diversity of microvessel density(MVD) in tumor tissue was detected by immunohistochemistry. RESULTS: Middle and high dose thalidomide could significantly inhibit the growth of OVCAR-3 human tumor in nude mice(P<0.05,P< 0.01), but the effect on HCT-8 growth was not obvious. Thalidomide could dose-dependently lower MVD in tissues in all the tumors. CONCLUSION: The anti-tumor effect of thalidomide may be irrelevant to its anti-neovascularization.
