载脂蛋白E(ApoE)基因多态性与2型糖尿病(T2DM)血脂及其脑梗死并发症的关系研究

摘    要:目的分析并研究载脂蛋白E基因多态性与2型糖尿病血脂及其脑梗死并发症的关系。方法从2012年5月—2014年5月期间该院收治的2型糖尿病患者中选取100例作为临床研究对象,设定为试验组,其中50例患者并发脑梗死,50例患者无并发症;同时从全市范围内征集100例健康者作为对照组。两组实验者均采用聚合酶链反应-限制性片段长度多态性方法检测其Apo E基因多态性,通过基因多样性的检结果分析并研究其与2型糖尿病血脂及其脑梗死并发症的关系。结果对照组和试验组等位基因分布频率相差不大,差异无统计学意义(P〉0.05);对试验组的100例患者进行组间比较发现,患者低密度脂蛋白与胆固醇含量在携带E4等位基因的患者中最高,在携带E2等位基因的患者中最低,差异有统计学意义(P〈0.05)。且实验结果显示并发脑梗死的患者E4等位基因的携带率高,无并发脑梗死的患者E4等位基因的携带率低,差异有统计学意义(P〈0.05)。结论载脂蛋白基因多样性与2型糖尿病之间不存在必然的关系;2型糖尿病高脂血症及其并发脑梗死与E4等位基因的携带有关。

Study of the Relationship Between Apolipoprotein E(Apo E) Gene Polymorphism,Type 2 Diabetes(T2DM) Blood Lipid and Cerebral Infarction Complication

Abstract:Objective To analyze and study the relationship between apolipoprotein E gene polymorphism and type 2 diabetes blood lipid and complication of cerebral infarction. Methods 100 cases with type 2 diabetes admitted in our hospital from May2012 to May 2014 were selected as the clinical subjects and set as the experimental group, including 50 cases with cerebral infarction and 50 cases without. And 100 healthy controls selected from the city were set as the control group during the same period. Polymerase chain reaction-restriction fragment length polymorphism method was used to detect the Apo E gene polymorphism of the two groups. The detection results of genetic diversity were analyzed and the relationship between type 2 diabetes blood lipid and complication of cerebral infarction was studied. Results The distribution frequency of allele in the control group and the experimental group had no obvious difference, the difference was not statistically significant(P〉0.05); It was found by comparing 100 patients in the experimental group that patients carrying E4 allele had the highest low density lipoprotein and cholesterol content,while patients carrying E2 allele had the lowest, the difference was statistically significant(P 0.05). The experimental results showed that diabetic patients with cerebral infarction had high carrying rate of E4 allele, while those without cerebral infarction had low carrying rate of E4 allele, the difference was statistically significant(P〈0.05). Conclusion There is no necessary relationship between diversity of apolipoprotein gene with type 2 diabetes; type 2 diabetic hyperlipemia and cerebral infarction complication have certain relationship with the carrying of E4 alleles.
