小檗胺对视网膜母细胞瘤HXO-RB44细胞增殖的抑制作用

摘    要:目的:探讨小檗胺对视网膜母细胞瘤HXO-RB44细胞增殖和细胞周期的影响.方法:体外培养视网膜母细胞瘤HXO-RB44细胞,四甲基偶氮唑蓝(MTT)法测定0、2、4、8、16、32 mg/L小檗胺作用24、48和72 h后对HXO-RB44细胞增殖的抑制率,流式细胞仪检测0、4、8、16 mg/L小檗胺作用HXO-RB44细胞24h后的细胞周期.结果:随着小檗胺浓度的增加和作用时间的延长,其对HXO-RB44抑制率逐渐增加,作用24、48、72 h的IC50分别为25.26、10.94和6.25mg/L.流式细胞分析显示,与空白对照组比较,小檗胺可显著抑制细胞周期,使G2/M期细胞增多(P<0.01).结论:小檗胺在体外可抑制视网膜母细胞瘤HXO-RB44细胞的增殖,并可诱导G2/M期阻滞,可能成为一种有效的抗视网膜母细胞瘤药物.

Abstract:Objective: To investigate the effect of berbamine (BBM) on the proliferation and cell cycle of retinoblastoma HXO-RB44 cells. Methods: HXO-RB44 cells were cultured in vitro. The inhibitory effect of BBM on the cell proliferation was assayed by MTT method after being interfered for 24 h, 48 h and 72 h by 0, 2, 4, 8, 16 and 32 mg/L, respectively. The cell cycles were analyzed by flow cytometry (FCM) after being interfered 24 h by 0, 4, 8 and 16 mg/L BBM, respectively. Results: The cell proliferation was significantly inhibited by BBM in a dose-dependent manner (P < 0.05). Values of IC50 were 25.26, 10.94 and 6.25 mg/L for 24 h, 48 h and 72 h after treatment , respectively. The result of FCM analysis showed that BBM significantly inhibited the cell cycle, and increased the number of cells in G2/M phase compared with those of control group (P <0.01). Conclusion: Berbamine can inhibit the proliferation of HXO-RB44 cells and induce cell cycle arrest in G2/M phase, which suggests that BBM may be an effective anti- retinoblastoma drug.
