新型生物人工肝系统治疗急性肝衰竭犬后猪内源性逆转录病毒传播的实验研究

摘    要:目的:探讨猪肝细胞为基础的新型生物人工肝(BAL)系统体外灌流后及治疗急性肝衰竭(ALF)犬后猪内源性逆转录病毒(PERV)的传播。方法:构建新型BAL,应用BAL进行体外灌流6h。采用门腔分流及胆总管结扎切断术建立犬ALF模型,应用BAL治疗ALF犬6h。采用PCR检测灌流前猪细胞中PERV原病毒DNA和猪mtDNA,采用RT-PCR检测灌流前后中空纤维管内腔循环液和治疗前后犬血清PERV RNA。结果:猪细胞可检测到PERV原病毒DNA和猪mtDNA,灌流前后中空纤维管内腔循环液和犬血清PERV RNA均为阴性。结论:猪肝细胞为基础的新型BAL体外灌流和用于ALF犬治疗6h后未发现PERV传播。

Abstract:Objective: To study the transmission of porcine endogenous retrovirus after perfusion in vitro and treatment for acute liver failure canines using bioartificial liver system based on porcine hepatocytes. Methods:The novel bioartificial liver system (BAL) was constructed. The hepatocyte suspensions were circulated in BAL system for 6h. ALF was induced by end-side portocaval shunt combined with common bile duct ligation and transection. BAL was used to treat acute liver failure canines for 6h. PCR was used for detection of PERV of porcine hepaocytes. RT-PCR was used for detection of PERV RNA of circulated liquid in the hollow fiber calibers and serum of canines pre-circulation to post-circulation. Results:The results showed that PERV was detectable in porcine hepaocytes and PERV RNA was not detectable in the circulated liquid and sera of the canines pre-circulation and post-circulation. Conclusion:The spread of PERV had not been found after circulation of 6h in vitro and treatment of 6h for ALF canines using the novel BAL system based on porcine hepatocyte.
