原子力显微镜对正常细胞、肿瘤细胞细胞膜表面形态结构的观察

目的 寻找原子力显微镜（AFM）观察培养细胞获得理想图像的样本制备方法；运用AFM探寻正常细胞膜与肿瘤细胞膜表面结构的异同。方法 摸索培养细胞制备成AFM扫描用标本的方法并进行扫描。结果 2％福尔马林溶液固定标本和枸橼酸盐缓冲液冲洗标本的细胞膜表面都很干净，5％葡萄糖溶液冲洗后标本图像模糊成比。正常Fb细胞膜由较规则的突起与凹陷构成。Ma-Fb细胞膜隆起结构大小相差悬殊，形态多样而且Z轴方向上高度明显高于Fb细胞。Hela细胞和Mcf7细胞均由形状不规则的突起与凹陷构成。结论 2％福尔马林溶液和枸橼酸盐缓冲液适用于AFM标本的制备：正常细胞膜表面隆起与凹陷结构的形态比肿瘤细胞膜规则，大小也较均匀；同种细胞恶变化细胞膜隆起结构形态多样而且体积明显增大；细胞种类不同，细胞膜表面结构也不同。

Observation of the membrane of normal cells and tumor cells by AFM

Objective Objective To find out the best method of observation of culture cells under AFM.Normal cells and tumor cells were observed by AFM in order to find out the difference between their membrane structures.Methods We chose Fb cell,Hela cell,MCF7 cell,Ma-Fb cell.To find the AFM specimen preparation and obtain their AFM pictures.Results The AFM images of the cells dealt with 2% formaldehyde solution and citrate buffer are clear,the AFM pictures of the cells with 5% dextrose solution are illegibility.The membrane configurations of Fb cell and Ma-Fb cell are defferent observably.Other two kind's cells membrane structures are abnormally.Conclusion 2% Formaldehyde solution and citrate buffer are fit for the preparation of AFM specimens.The membrane configurations of normal cell and tumor cell are obviously different.
