摘    要:目的:探讨半边旗提取物5F对人高转移卵巢癌细胞HO-8910PM细胞周期的影响及其作用机制.方法:以MTT法检测5F对高转移卵巢癌细胞HO-8910PM细胞增殖的影响;流式细胞术分析5F对HO-8910PM细胞周期的影响;Western blot法分析NF-κB(P65)、FAK的表达及FAK酪氨酸磷酸化水平.结果:不同浓度的5F分别处理细胞24 h后,对HO-8910PM细胞增殖有明显的抑制作用;流式细胞术分析表明5F使G0/G1期的细胞减少,阻断细胞于G2/M期;同时,5F可使NF-κB(P65)的表达下调,上调FAK的表达,并降低FAK酪氨酸磷酸化水平.结论:5F对HO-8910PM细胞周期的影响与NF-κB(P65)表达下调及FAK的表达上调有关.

Abstract:OBJECTIVE: To study the effect and its possible mechanism of 5F from Pteri semipinnata L on the cell cycle of human highly metastatic ovarian carcinoma HO-8910PM cells. METHODS: MTT assay was used to examine the effect of 5F on proliferation of HO-8910PM cells after 24 hours treatment; The cell cycle was assessed by flow cytometry (FCM); The expression level of NF-kappaB (p65) FAK and the level of phosphorylated FAK were assessed by Western blot analysis. RESULTS: 5F from Pteris semipinnata L. could inhibit the proliferation of HO-8910PM cell and block the cell cycle at G2/M phase. The expression level of NF-kappaB (P65) protein decreased obviously in HO-8910PM cells treated with 25 - 100 micromol/L 5F for 24 hours, and the effect appeared in a dose-dependent manner. 5F up-regulated significantly the expression of FAK, down-regulated the level of phosphorylated FAK. CONCLUSION: The effect on cell cycle of 5F from Pteri semipinnata L is involved in the expression of NF-kappaF(p65), FAK and the level of phosphorylated FAK.
