摘    要:目的 探讨慢性氟中毒、砷中毒及联合中毒对小鼠肾皮质氧化应激损害程度及其异同 ,为氟、砷中毒的防治提供科学依据。方法 昆明种小鼠 1 2 0只 ,随机分成对照、氟、砷和氟砷联合染毒 4组 ,分别自由进食普通、高氟、高砷和高氟高砷饲料 ,分别于 6个月和 1 2个月取材 ,检测肾皮质丙二醛 (MDA)和脂质过氧化物 (LPO)含量、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶 (GSH PX)、琥珀酸脱氢酶 (SDHase)和葡萄糖 6 磷酸酶 (G 6 Pase)活力。结果 与对照组比较 ,3种染毒模式在 6和 1 2个月观察期均引起肾皮质MDA含量增加 (P <0 0 1 ) ,GSH PX 活力在 6个月时均下降 (P <0 0 1 ) ;仅见氟组在 1 2个月时恢复正常 ,而砷和氟 +砷组仍维持低水平 (P <0 0 1 ) ;2个观察时点的SOD活力均下降 (P <0 0 1 )。此外 ,1 2个月的GSH、SDH和G 6 Pase均下降 (P <0 0 1 ,P <0 0 1 ,P <0 0 5)。结论 慢性氟、砷和氟砷联合染毒均能引起小鼠肾皮质氧化应激作用增强 ,氟、砷 2毒物对在引起肾皮质LPO损害作用方面无明显交互作用

Abstract:Objective To investigate the effects of oxidative stress in kidney of mice under the condition of chronically exposed to fluoride ,arsenic or both fluoride and arsenic respectively.Methods 120 mice were randomly separated into four groups inclu d ing fluoride,arsenic,fluoride with arsenic,and control groups,which were fed dif ferent diets supplemented with fluoride,arsenic,fluoride with arsenic,or none,re spectively.After six and twelve months,the animals were sacrificed and a series of biochemical indices were detected in kidney cortex.Results Compared with control group,the concentration of malona ldehyde(MDA)in kidney cortex increased significantly in all three models either in 6 months or 12 months intoxicated mice(P<0 01);Activities of glutathione peroxidase(GSH-P X) constantly decreased both in 6 months and 12 months(P <0 01) except for single increase to control level in fluoride group of 12 months ;Activity of supperoxide dismutase(SOD) decreased significantly in all three mod els e ither in 6 months or 12 months-observation(P<0 01).Furthermore,concentrati ons of glutathion(GSH),succinate dehydrogenase(SDH) and glucose-6-phosphatase( G6P) decreased in 12 months intoxicated mice (P<0 01,P<0 01 and P< 0 05 respectively).Conclusion Chronically exposing fluoride,arsenic or both fluoride a nd arsenic can increase oxidative stress in mouse kidney cotex and it seems that the interaction between fluoride and arsenic is not significant besides.
