人眼眶前脂肪细胞的培养和诱导分化及鉴定

目的近来研究显示,人眼眶脂肪组织中的前脂肪细胞可能参与多种疾病的发病过程。文中建立人眼眶前脂肪细胞原代培养模式,对其进行培养、诱导分化及鉴定,以便深入了解人眼眶前脂肪细胞的生物学特性。方法从18～56岁成人眼眶脂肪组织中分别以组织块法及酶消化法分离得到前脂肪细胞,进行形态学观察,成脂诱导分化,并对其进行鉴定。结果 2种方法均可获得前脂肪细胞,细胞为长梭形,呈"S"形生长,可成脂分化,且油红O染色阳性。结论从人眼眶脂肪组织中可分离出纯度高、增殖快、成脂分化率高的前脂肪细胞。
