摘    要:目的:探讨在肝脏缺血再灌注损伤中,应用rhGH(重组人生长激素)后,对肝细胞能量代谢的影响。方法:(1)用Pringle s法建立80只Sprague Dawley(SD)大鼠肝脏缺血再灌注损伤模型,实验组40只,对照组40只,另10只作空白对照。实验组皮下注射rhGH针剂0.2 U·100g~(-1)·d~(-1)共7 d,对照组分别给予等量的生理盐水。(2)分别于术后第1天、第7天、第14天、第28天取肝组织用高效液相色谱法检测ATP、ADP、AMP,并计算EC(能荷)。结果:rhGH组和对照组不同时点的EC水平的组间同期比较示:在1 d、28 d无显著差异(P>0.05),7 d、14 d有显著差异(P<0.05);组内14 d与28 d比较,无显著差异(P>0.05),其余各时点均有显著差异(P<0.05)。结论:rhGH能改善肝脏缺血再灌注损伤时肝细胞能量代谢状态。

Abstract:Objective:To investigate variation of hepatocelluar energitic metabolism after practicing rhGH in ischemic reperfu- sion of rat liver.Methods:Ninety male SD rats,which weight 200-300g,were randomly divided into:rhGH group(n=40), control group(n=40)and blank control group(n=10).In rhGH group,the rhGH was administered to rats via hypodermic in- jection(0.2UI/100g SD rats weight,once daily)seven days after establishment of the model of hepatic ischemic reperfusion,in control group rats were treated as rhGH group except for rhGH which was replaced by normal sodium.Then the rhGH group and the control group were further divided into 4 groups equally according to the period of ischemic reperfusion,as 1 day,7days ,14days,28days,following establishment of the model,the level of energy charge(EC)was detected.Results:Increase of EC in rhGH group were faster than those of control group,there was significantly difference on the 7~(th)day and 14~(th)day(P<0.05).Conclusion:Injecting rhGH after ischemic reperfusion of rat liver,rhGH can improve level of hepatocelluar EC.
