PLGA纳米载体介导的端粒酶hTERT反义核酸对裸鼠肝癌移植瘤的影响

摘    要:目的通过以PLGA纳米粒作为端粒酶hTERT反义核酸的载体,促进反义核酸对肝癌瘸组织的抑制。方法以HepG-2肝癌细胞和裸鼠建立肝癌移植瘤实验模型;观测瘸重和成瘤率;流式细胞仪检侧肿瘤细胞凋亡及细胞周期的特点。结果与对照组相比,治疗组肿瘤细胞凋亡数明显增加,细胞增殖受到抑制,肿瘤生长缓慢,出瘤时间推迟。结论以PLGA纳米粒作为端粒酶反义核酸的栽体,能显著延缓裸鼠肝癌移植瘤的发展,产生明显的疗效。

Abstract:[Objective] To improve inhibition of liver tumor through telomerase antisensenueleie acids carried by PLGA nanopartieles. [Methods] To establish animal model with transplanted liver tumor by using HepG-2 cells and athymie mouse. To investigate weight and emerging rate of tumor. To inspect apoptosis and cell cycle of tumor cell by fllow eytometry. [Result] Comparing with control group, apoptosis turnout cells increased in treated group and cell nndtiplieation was inhibited, growth of tumor slowed. [Conclusion] Telomerase antisensenueleie aeids earfled by PLGA nanopartieles could postpone development of liver tumor and create evid ctlrative effect.
