脑胶质瘤的化疗敏感性研究

摘    要:目的应用MTT比色法比较六种抗肿瘤药物对脑胶质瘤细胞的抑制作用,为临床化疗筛选药物、制定化疗方案提供指导。方法应用MTT比色法对62例脑胶质瘤新鲜标本进行化疗药物敏感性研究。结果六种药物敏感率经卡方检验有明显差别,由高至低依次为:Vm-26、Fotemustine、BCNU、CBP、DDP、Vp-16。病人性别对药物敏感率无影响,年龄、术前机能状态KPS评分对大多数药物敏感率无影响,肿瘤病理分级只对部分药物敏感率有影响。结论MTT比色法是一种快速、准确检测体外药物敏感性的方法。Vm-26抑制肿瘤细胞增殖作用较强,优于Fotemustine、BCNU、CBP、DDP、Vp-16。

Abstract:Objective To compare the effect of 6 antineoplastic agents on suppressing cerebral glioma cells in vitro with colorimetric MTT assay, so as t o supply some instructions for selecting chemotherapeutic plan for individual pa tient. Methods 62 neurosurgical biopsies were plated in 96-well tissue cul ture plates and incubated for 72 h with 6 different anti-cancer agents, and 3 di fferent concentrations of each agent were tested. The MTT dye was then added to the wells, and the resulting formazan precipitate was solublized with 10%SDS. Th e spectrophotometric absorbency (measured at 570 nm) of control and experimental wells was used to calculate the cytotoxicity index (CI). Results 6 agents ' chemosensitivity was significantly different. Rank Test listed them from high to low: Vm-26, Fotemustine, BCNU, CBP, DDP, Vp-16. Patient age, sex and Karnofsk y Performance Status (KPS) did not prove to be significantly correlated with the most agent chemosensitivity in vitro by statistics. Pathological grade was corr elated with chemosensitivity of the part of agent. Conclusion The MTT assa y is a rapid method of screening antineoplastic agent against gliomas for cytoto xicity. The gliomas cell is more sensitive to Vm-26 in vitro than to Fotemustine , BCNU, CBP, DDP, Vp-16.
