温郁金对VEGF和MVD在人胃癌裸小鼠移植瘤中表达的研究

摘    要:目的研究温郁金对人胃癌裸鼠移植瘤生长的抑制作用和对血管内皮生长因子(VEGF)的表达和微血管密度(MVD)的影响.方法采用人胃癌SGC7901细胞株种植裸小鼠皮下建立胃癌移植瘤模型.自肿瘤种植后,待肿瘤生长到2 mm时开始给药,每日2次,连续7周,每周测量瘤体的大小.第7周处死动物,测定肿瘤重量,免疫组化ElivisionTM plus法测定VEGF及肿瘤内MVD.结果温郁金治疗组瘤重和体积明显低于对照组,VEGF表达的程度和MVD计数明显低于对照组.结论温郁金对人胃癌裸鼠移植瘤的生长具有明显的抑制作用,可下调瘤灶中VEGF的表达,减少肿瘤灶内的MVD.

Abstract:Objective Inhibition effect of common turmeric (wenyujin) on the growth of tumor transplanted from Human Gastric Carcinoma to athymic mouse and its effect on the expression of vascular endothelial growth factor and microvessel density. Methods A mouse tumor model was set up by implanting SGC-7901 human gastric cancer cells under the skins of nude mice.Twelve experimental mice were divided into 2 groups,6 in common turmeric group and other 6 in control group. When the diameter of the tumors grew up to 2mm, the experimental group was administrated with a dose of 0.3ml vaporized distilled common turmeric (the concentration was 250%) twice a day for 7 weeks, while the control group was administrated with the same dose of saline meanwhile. During experiments, the tumors' size was measured at a fixed time each week. After 7 weeks, the nude mice were killed and the tumors were taken out and weighed to be examined by using immunohistochemical staining to study the expression of vascular endothelial growth factor and microvessel density. Results The weight and size of the tumors in the experimental group was less than that of the control group significantly, and so was the expression of vascular endothelial growth factor and microvascular density. Conclusion Common turmeric show a significant inhibitory effect not only on the growth of the tumors but on the expression of the vascular endothelial growth factor and the microvascular density.
