CEP65在肝癌中的表达及其对肝癌细胞迁移、侵袭的影响

目的：探讨肿瘤及胚胎组织特异表达蛋白65（cancer and embryo expression protein 65，CEP65）在肝癌组织中的表达，分析siRNA干扰CEP65后对肝癌细胞迁移及侵袭的影响。方法：免疫组化法检测肝癌组织中CEP65的表达情况；Western blot法检测CEP65在肝癌细胞中的表达水平，并通过siRNA干扰CEP65后Tran-swell实验检测敲低CEP65表达后对肝癌细胞迁移及侵袭的影响。结果：CEP65在肝癌组织中的表达明显高于癌旁组织；肝癌组织中CEP65高表达与TNM分期、门静脉侵犯及Edmondson分级显著相关；高表达CEP65患者5年生存率明显低于CEP65低表达组；siRNA干扰肝癌MHCC－97H细胞CEP65的表达后可显著抑制其迁移及侵袭。结论：CEP65在肝癌组织中的表达明显升高，并促进肝癌细胞迁移及侵袭。

The expression of CEP65 in hepatocellular carcinoma and its significance in inhibiting mi-gration and invasion

Objective:To investigate the expression of CEP65 in hepatocellular carcinoma(HCC)patients and ex-plore its role in migration and invasion of HCC cells through RNA interference.Methods:The expression of CEP65 in HCC tissues was assessed by immunohistochemistry.The expression level of CEP65 was determined by Western blot. The effect of CEP65 knockdown by siRNA on migration and invasion of HCC cell was observed by Transwell assay.Results:CEP65 expression was significantly highly expressed in HCC tissues compared to tumor-adjacent tissue.Its high expression was significantly associated with TNM stage,portal vein infiltration and Edmondson degree.Patients in the high CEP65 group had a lower 5 -year survival than those in low CEP65 group.Down-regulated of CEP65 by siRNA could inhibit cell migration and invasion in MHCC-97H cell.Conclusion:The expression of CEP65 in HCC increased significantly,and it contributes to the migration and invasion of HCC cells.
