FOXC2蛋白在食管鳞状细胞癌组织中的表达及与预后的相关性

目的：叉头框 C2（FOXC2）在多种肿瘤中表达，并与肿瘤的转移和增殖关系密切，但其在食管鳞癌中的表达以及作用未见报道，本研究旨在探讨 FOXC2蛋白在食管鳞癌组织中的表达及与预后的相关性。方法：应用实时定量 PCR 法以及 Western blot 法检测 FOXC2在129例食管鳞癌组织中的表达情况，并且分析其表达水平与患者性别、年龄、肿瘤位置、肿瘤大小、分化程度、T 分期、淋巴结转移等临床病理之间的关系，研究FOXC2对患者预后的影响。结果：PCR 证实在 mRNA 水平 FOXC2在食管鳞癌组织中表达水平均高于其对应的癌旁组织（P ＝0．005）。免疫组化结果表明 FOXC2在食管鳞癌中的阳性率为60．47％，与患者的肿瘤大小（P ＜0．001）、T 分期（P ＝0．008）、淋巴管转移（P ＝0．005）关系密切，而与患者的年龄、肿瘤位置等没有统计学意义；FOXC2阳性表达的患者5年生存率显著低于 FOXC2阴性表达的患者（P ＝0．009）；同时，多因素分析显示 FOXC2是食管癌患者预后的独立因素。结论：FOXC2高表达于食管鳞癌组织中，并且发现其与食管癌的转移密切相关，其对食管鳞癌患者的预后预测有重要意义，可能成为食管鳞癌诊断、治疗的新标志物。

The relationship between FOXC2 expression and prognosis in esophageal squamous cell carcinoma

Objective:To investigate the expression and effect of FOXC2 in esophageal squamous cell carcinoma (ESCC).Methods:The expression of FOXC2 was investigated in ESCC tissues and matched adjacent tissues.Fur-thermore,the relationship between expression level of FOXC2 and clinical features,including gender,age,tumor loca-tion,tumor size,differentiation,T stage and lymph node metastasis,was analyzed.The relationship of FOXC2 expres-sion influenced on prognosis of patients was explored.Results:Expression of FOXC2 was significantly higher in ESCC tissues than matched adjacent tissues by PCR (P =0.005).Positivity of FOXC2 staining was 60.47% in ESCC,and there was significant association between FOXC2 and tumor sizes (P <0.001),T stage (P =0.008),lymph node metastasis (P =0.005).The 5 -year survival rate of patients with expression of FOXC2 was lower than that of pa-tients with negative expression.Furthermore,expression of FOXC2 was an independent prognostic factor for overall survival.Conclusion:In conclusion,this study indicated that expression of FOXC2 was significant higher in ESCC tis-sues,and it was closely related to the metastasis of tumor.FOXC2 may be a new marker for diagnosis and treatment of esophageal squamous carcinoma.
