玉簪花的色谱鉴别和含量测定研究

摘    要:目的:建立玉簪花的鉴别方法及其山柰素含量的测定方法。方法:鉴别采用薄层色谱法;含量测定采用高效液相色谱法,应用Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.4%磷酸溶液(60∶40)为流动相,流量为1 mL.min-1,检测波长为360 nm,柱温为30℃。结果:薄层色谱分离好,斑点清晰,阴性对照无干扰;山柰素在0.05~0.30μg范围内具有良好的线性关系,r=0.999 9,平均回收率为100.26%、RSD为1.69%(n=9)。结论:该方法灵敏、准确、专属性强,重现性好,可用于玉簪花的质量控制。

Abstract:Objective: To establish a method for identification of Hostae Flos and determination of kaempferol in it. Methods: The TLC method was used to distinguish Hostae Flos and HPLC for determination of kaempferol. The Diamonsil Cis (4. 6 mm .250 mm,5 μm) column was used and methanol-0. 4% phosphoric acid solution(60: 40) as mobile phase with flow rate of 1 mL. min . The detection wavelength was 360 nm and column temperature was 30℃. Results : The TLC identification was highly specific and spot of ofkaempferol was clear and there is no interference in negative comparison. Kaempferol showed a good linearity in a concentration range of 0. 05-0. 30 μg (r =0. 999 9) . The average recovery was 100. 26% and RSD was 1.69% (n =9). Conclusion: This method was sensitive and accurate, in strong specificity and in good repeatability,it can be used for the quality control of Hostae Flos.
