钨磷酸盐联合表阿霉素抑制K562细胞增殖

摘    要:目的:探讨低浓度的钨磷酸盐(K6P2W18O62]·14H2O)对表阿霉素抑制K562细胞增殖的影响与机制。方法体外培养K562细胞,不同浓度钨磷酸盐和表阿霉素处理后,WST-1还原法检测细胞增殖、流式细胞术检测细胞内表阿霉素的相对含量和活性氧值。结果钨磷酸盐(15μmol/L)与阿霉素(2.5、5和10μmol/L)的联合指数分别为1.62和1.29。结论两者具有协同抗肿瘤效应;钨磷酸盐可促进阿霉素内化入细胞并降低细胞内活性氧水平;钨磷酸盐可能通过促进表阿霉素内化入细胞而发挥协同抗肿瘤的作用。

Abstract:Objectove To probe the effect and its mechanism of low concentration of K6 P2 W18 O62 ]·14 H2 O on epi-rubicin inhibiting cell proliferation of K562 . Methods K562 cell was cultured in vitro and processed in different concentration K6P2W18O62]·14H2O and epirubicin. The cell proliferation was detected by WST-1 reduction meth-od. The epirubicin relative amount intracellular and reactive oxygen species value were detected by flow cytometry. ResuIts The combination index of K6P2W18O62]·14H2O(15 μmol/L)and epirubicin(2. 5 μmol/L,5 μmol/L and 10 μmol/L)was 1. 62 and 1. 29,respectively. concIusoon Tungsten phosphate could promote epirubicin into cells and reduce the active oxygen levels intracellular. Tungsten phosphate can get into cells through epirubicin and has effect of synergistic antitumor.
