中波紫外线照射对HaCaT细胞p53mRNA和p53蛋白表达的影响

摘    要:目的:研究中波紫外线(UVB)照射对永生化角质形成细胞株-HaCaT细胞的p5 3mRNA和p53蛋白表达的影响.方法:以一定剂量UVB照射HaCaT细胞,分别用RT-PCR法和Western blot方法检测照射后不同时间点的p5 3mRNA和p5 3蛋白的表达水平;同时检测不同剂量UVB照射后同一特定时间点p5 3mRNA和p53蛋白的表达水平.结果:30mJ/cm^2的UVB照射后HaCaT细胞的p5 3mRNA和p5 3蛋白表达逐渐增加,4h达到峰值,后逐渐恢复至照光后24h接近正常值;不同剂量UVB照射4h后,p53mR-NA和p53蛋白表达随剂量增加而增加.结论:HaCaT细胞p5 3mRNA和p5 3蛋白表达与UVB照射存在一定的时间和剂量关系.

Abstract:Obiective To investigate the effects of UVB irradiation on p53mRNA and p53 protein expression of HaCaT cells. Methods HaCaT cells were cultured and irradiated with certain dosage of UVB. p53 mRNA and p53 protein expressions on different time and of different dosages on certain time were measureded by RT-PCR and Western blot. Results p53mRNA and p53 protein expression of HaCaT ceils irradiated with 30mJ/cm^2 UVB increased gradually and reach the peak at the 4 hours after irradiation then deceased close to normal level at 24 hours after irradiation.p53 mRNA and p53 protein expression increased with the increasing dosages on 4 hours after irradiation. Conclusion p53 mRNA and p53 protein expression of HaCaT cells were time-dependent and dosage-dependent with UVB irradiation.
