摘    要:作者试用媒蚊离体吸血法,观察抗间日疟红内期原虫单克隆抗体对配子体的感染力及其在蚊体内发育的影响,获得成功。受试的五株杂交瘤细胞克隆或亚克隆腹水抗体中,1D_6—G_7对配子体的感染力具有明显抑制作用,抑制率高达84.6%。另有三株也对配子体具不同程度的抑制效果。提示此类单克隆抗体在阻断间日疟传播方面将有重要的应用研究价值。

Abstract:Using hybridoma technique, a panel ofmonoclonal antibodies against erythrocyticstages of Plasmodium vivax has been pro-duced and reported elsewhere. In order toidentify whether these McAbs have protectivesignificance, we chose five of them withbroad-spectral IFA reactions (1D_6-G_7; 6H_7;7A_7; 6F_7 and 1D_6-H_(10)) for this study. McAbs were fed to Anopheles stephensitogather with the patient's gametocyte-con-taining blood through the membrane feeders.The effect of the test McAbs in the bloodmeal was compared with that of both posi-tive and negative controls and measured bysubsequent examination of the mosquitoesfor the presence of oocysts and aporozoites. Six experiments were successfully carriedout during two epidemic seasons in 1986-1987. Results of these membrane feedingexperiments demonstrated that four McAbsinhibited gametocyte infectivity to differentextent as compared with the negative con-trols. Among them, 1D_6-G_7,a subclone,was shown to be the most effective one.Under the same experimental conditions, thepercentage of mosquitoes infected in 1D_6-G_7, chloroquine, SP_(2/0) ascites and untreatedgroups was 20. 0%, 60.0%, 70.0% and 57.7%respectively; the average number of oocystsper positive gut dissected in the correspond-ing groups was 4. 1 (P<0.01) 13. 8, 38. 5 and24.7 respectively. According to the meannumber of oocysts, the inhibitory rateinduced by 1D_6-G_7 was 84. 6%. Since McAb 1D_6-G_7 has apparentlysuppressive effect on gametocyte infectivityand its development within mosquito vectorsshown above, it may play an important rolein the further studies on the transmission-blocking immunity in Plasmodium v vax.
