非小细胞肺癌EGFR基因突变与E-cadherin、Vimentin表达的相关性

摘    要:目的:探究非小细胞肺癌患者肺组织中E-cadherin、Vimentin表达与EGFR突变的关系。方法:收集在本院进行治疗的非小细胞肺癌患者104例作为研究对象,采用PCR检测患者肿瘤组织中EGFR基因突变情况,采用免疫组化法检测肺组织中E-cadherin、?Vimentin表达情况,分析EGFR基因突变、E-cadherin、?Vimentin与患者临床参数的关系;Pearson相关系分析E-cadherin、?Vimentin相关性;Logistic回归分析影响非小细胞肺癌患者EGFR突变的危险因素。结果:非小细胞肺癌患者EGFR突变61例。非小细胞肺癌患者EGFR基因突变与患者性别、肿瘤组织类型、患者吸烟有关(P<0.05)。与EGFR基因野生型患者相比,EGFR基因突变者肺组织中E-cadherin表达显著升高,Vimentin表达显著降低(P<0.05)。E-cadherin、?Vimentin与非小细胞肺癌患者肿瘤分期相关(P<0.05)。Pearson相关性分析显示,非小细胞肺癌患者E-cadherin与Vimentin呈显著负相关(r=-0.483,P<0.05)。结论:非小细胞肺癌患者E-cadherin高表达,Vimentin低表达与EGFR突变患者耐药有关。

Abstract:Objective: To explore the relationships between the expressions of E-cadherin and Vimentin and EGFR mutation in lung tissues of drug-resistant patients with non-small cell lung cancer. Methods: 104 patients with non-small cell lung cancer who were treated in our hospital were collected as the study subjects, the mutation of EGFR gene in tumor tissues was detected by PCR, the expressions of E-cadherin and Vimentin in lung tissue were detected by immunohistochemistry, the relationships between EGFR gene mutation, E-cadherin, Vimentin and clinical parameters were analyzed; Pearson correlation coefficient was used to analyze the correlation between E-cadherin and Vimentin; and Logistic regression analysis was used to analyze the risk factors of EGFR mutation in patients with non-small cell lung cancer. Results: 61 patients with non-small cell lung cancer had EGFR mutation. The mutation of EGFR gene in non-small cell lung cancer (NSCLC) patients with chemotherapy resistance was related to gender, tumor tissue type and smoking (P < 0.05). Compared with wild-type patients with EGFR gene, the expression of E-cadherin in lung tissue of EGFR gene mutation was significantly increased, and the expression of Vimentin was significantly decreased (P < 0.05). E-cadherin and Vimentin were correlated with the stage of non-small cell lung cancer (P < 0.05). Pearson correlation analysis showed that E-cadherin was negatively correlated with Vimentin in non-small cell lung cancer (r=-0.483, P < 0.05). Conclusions: High expression of E-cadherin and low expression of Vimentin in patients with non-small cell lung cancer are associated with drug resistance in patients with EGFR mutation.
