苦参碱对人增生性瘢痕成纤维细胞增殖的影响

摘    要:目的:探讨苦参碱(Matrine,简称Mat)的药理活性,研究苦参碱对人增生性瘢痕成纤维细胞增殖的抑制作用,为皮肤损伤后出现的增生性瘢痕的治疗提供理论支持及实验依据。方法:将不同浓度梯度Mat作用于体外培养的增生性瘢痕成纤维细胞,测MTT反应的A。值及LDH值,利用通过流式细胞仪检测其细胞周期、DNA相对含量,用免疫组织化学检测Mat用药前后的细胞核内增殖性抗原(pr01iferatingcell nuclear antigen,PCNA)的表达。结果:增生性瘢痕成纤维细胞在一定范围内呈剂量依赖性增殖抑制,但LDH值无明显改变;DNA相对含量无明显变化,PCNA表达逐渐减弱。结论:Mat在体外能明显抑制增生性瘢痕成纤维细胞的增殖而不引起细胞坏死性改变,但对DNA含量无明显影响。

Abstract:Objective To investigate the effects of matrine (Matrine,referred to as Mat) pharmacological activity,and study inhibitory effect of matrine on hyperplastic scar fibroblasts proliferation, provide theoretical support and experimental basis for the treatment of hyperplastic scar after skin injury. Methods In vitro different concentration gradient of Mat were used to cultured hypertrophic scar fibroblasts,then detected A^0 value and LDH value of MTT reaction,etected the cell cycle and the relative content of DNA by flow cytometry, before and after medication examined expression of nuclear Proliferating Antigen (proliferatingcell nuclear antigen, PCNA) by immunohistochemistry. Results In a certain range,hypertrophic scar fibroblasts were induced dosedependently proliferation inhibitory,but the LDH value did not change significantly.Relative content of DNA had no obvious change,expression of PCNA decreased. Conclusion Mat could significantly inhibit proliferation of hypertrophic scar fibroblasts without causing cell necrotic changes in vitro, but Mat had no obvious effect on the content of DNA.
