外源性NGF对脑脊液中氨基酸含量的影响

摘    要:目的 观察外源性神经生长因子(NGF)对实验动物脑脊液中氨基酸含量的影响,用以探讨NGF对脑神经细胞生物效应的作用机制。方法 应用反相高效液相色谱(HPLC)荧光检测方法,测量经不同途径(Vi、M、鼻饲)供给外源性NGF1周后,家兔脑脊液中氨基酸含量的变化。结果 试验组脑脊液中的兴奋性氨基酸(AST、Glu)明显减少(P<0.01),抑制性氨基酸(Ala、Gly)差异不显著(P>0.05)。而营养性氨基酸,特别是具有生物效应的His、Ser、TyT等均有明显增加(P<0.05)。结论 NGF进入体内可分解成小分子,并可通过血脑屏障调节脑内氨基酸浓度和神经细胞功能,达到保护和促进脑细胞生长的作用。

Abstract:Objective To content of amino acid in cerebrospinal fluid of experimental animals affected by exogenetic NGF has been observed,which can be used to investigate the mechanism of biological effect of brain nerve cells by NGF.Methods High Performance Liquid Chromatography (HPLC) had been used to determine the content of amino acid in rabbit cerebrospinal fluid after one week feeding of exogenetic NGF.Results The content of the excitatory amino acid of control group has been remarkably reduced ( P <0 01), while the difference of inhibitory amino acid can not be seen and nutritional amino acid,especially the biological His,Ser and Ty T have been tremendously increased.conclusion After entering the body,NGF had been disassembled into small molecules.They can enter the brain through BBB(blood-brain-barrier) to regulate the content of amino acid and nerve cells,which can protect the nerve cells and promote their growth.
