摘    要:目的建立高效液相-电雾式检测器(HPLC-CAD)方法测定远志药材中细叶远志皂苷的含量。方法色谱柱为Ultimate XB-C18柱(250mm×4.6mm,5μm);柱温25℃;流动相为甲醇-0.3mL·L~(-1)磷酸溶液(60︰40);流速1mL·min~(-1);进样量10μL;电雾式检测器,雾化温度25℃,采集频率10Hz。结果细叶远志皂苷峰面积与质量浓度线性回归方程:Y=4.295 4 X-0.135 8,r=0.999 9(n=6),线性范围为1.0~10.0mg·mL~(-1);平均回收率为101.4%。结论该方法高效、快速、准确,可作为药物及制剂中细叶远志皂苷的含量测定方法。

Determination of tenuifolin in Radix polygalae by HPLC-CAD
Abstract:Objective To establish a high performance liquid chromatographic with charge aerosol detection (HPLC-CAD) method for the determination of tenuifolin in Radix polygalae .Methods Ultimate XB-C18 column(250 mm × 4 .6 mm ,5 μm) was used in the study and the column temperature was 25 ℃ ,and methanol-0 .3mL?L -1 phosphoric acid (60︰40) solution as the mobile phase with a flow rate at 1 mL?min-1 ;the injection volume was 10 μL ;CAD :nebulization temperature was 25 ℃ and collection frequency was 10 Hz .Results The equation of linear regression was Y=4 .295 4X-0 .135 8 ,r=0 .999 9(n=6) ,linear range was 1 .0 10 .0 mg ? mL -1 ,and the average recovery was 101 .4% .Conclusion The method is accurate ,rapid and high sensitive , which can be used in the quantitative determination of tenuifolin in protein pharmaceuticals .
