葡萄酒协同洋葱对大鼠痛风症作用的研究

摘    要:目的通过尿酸钠(monosodium urate monohydrate,MSU)晶体诱导炎症动物模型,探讨红酒泡洋葱对急性痛风性关节炎(acute gouty arthritis,GA)的治疗作用。方法选用48只健康雄性SD大鼠,随机分为6组:空白组,模型组,阳性组,葡萄酒协同洋葱低剂量组,葡萄酒协同洋葱中剂量组和葡萄酒协同洋葱高剂量组。采用MSU晶体制备大鼠GA模型。空白组和模型组给予生理盐水,阳性组给予雷公藤多苷片,低、中、高剂量组分别给予不同剂量的葡萄酒+洋葱,检测血清中UA、IL-1β和CAT活性水平。取踝关节滑膜,观测其病理变化。结果低剂量(3mg·kg-1)能够显著降低GA模型大鼠的踝关节肿胀度及白细胞介素浓度(P<0.01),提高CAT活性(P<0.01);低、中(6mg·kg-1)剂量能够显著降低血清中尿酸的含量(P<0.01);尿酸钠所致痛风症大鼠的关节滑膜病变显著,经给药后各组大鼠的关节滑膜病理情况均有所缓解。结论葡萄酒泡洋葱可降低MSU晶体诱导的GA大鼠血清中尿酸及白细胞介素的水平,增强过氧化氢酶活性,对关节滑膜有保护和修复作用,其机制可能与抗氧化作用有关。

Study on wine combined with onion in the treatment of acute gouty arthritis
Abstract:Objective To study the therapeutic effect of wine combined with onion on acute gouty arthritis (GA) in the animal model of sodium urate (monosodium urate monohydrate ,MSU) crystal‐induced inflammations .Methods 48 healthy male SD rats were randomly divided into six groups :control group ,model group ,positive group ,low dose group ,middle dose group and high dose group .GA models were made by using MSU .Control group and model group were given normal saline ,positive group were given Tripterygiumwilfordii Glycosides Tablets ,and low dose group ,middle dose group and high dose group were given different doses of wine (onion) .Wine (onion) ,Tripterygiumwilfordii Glycosides Tablets or normal saline was given orally .The 1evels of UA , IL‐1βand CAT in plasma were determined with thiobarbituric acid (TBA) .The pathological changes of ankle synovial were ob‐served with microscope .Results Low dose of 3 mg · kg -1 wine (onion) significantly reduced ankle swelling and the contents of IL‐1β(P<0 .01) in GA rat ,increased the activity of CAT (P<0 .01);Low and middle (6 mg · kg -1 ) dose significantly de‐clined the level of serum UA (P<0 .01);Ankle synovial showed remarkable lesions in MSU‐induced GA rats ,treated with wine combined with onion ,and the pathology of ankle synovial were alleviated .Conclusion Wine combined with onion can reduce the levels of serum UA and IL‐1βin MSU‐induced GA rats ,enhance the activity of CAT ,indicating the therapeutic effect of protec‐ting and repairing the ankle synovial disease ,the mechanism of w hich may be related to the antioxidant effects .
