一氧化氮在犬马尾神经压迫后腰骶髓神经元凋亡发生中的作用

摘    要:目的探讨马尾神经压迫后NO含量变化在细胞凋亡中的作用。方法将24只健康家犬随机分为两组马尾神经压迫组(21只)假手术组(3只),马尾神经压迫组再分为7组即马尾神经压迫4h、8h、12h、24h、48h、72h和168h组(n=3)。水囊置于椎管内制成马尾神经压迫模型。检测NO含量变化,用TUNEL标记法标记凋亡神经元细胞。光镜(HE染色)和透射电镜观察细胞形态学改变。结果1假手术组细胞形态未见异常,压迫组光镜电镜结果均提示细胞发生凋亡。2假手术组未见TUNEL法标记凋亡神经元细胞,压迫组于12h即可见凋亡神经元细胞,24~48h神经元细胞凋亡最多。3压迫12h后,NO含量即明显增高,24~48h达到高峰,72h后开始下降。结论马尾神经压迫后犬腰骶髓神经元细胞凋亡发生早,持续时间长,NO含量增高与神经细胞凋亡指数间存在正相关。

Abstract:Objective:To observe the change of the content of NO during neuron apoptosis after the oppression of cauda equina nerve. Methods:24 healthy dogs were randomly divided into two groups: the oppressed cauda equina nerve group (21), and the fake operation group (3). The group of oppressed cauda equina never was randomly divided into seven groups, 3 dogs in each group. Cauda equina nerve was oppressed for 4h,8h,12h,24h,48h,72h and 168h. The model of oppressed cauda equina nerve was made by putting hyponome in spinal cord. The content of NO was detected, and the apoptosis was labeled by TUNEL. The morphological changes of neurons were observed by HE staining microscopic examination and transmission electron microscope. Results: The neurons in fake operation group were normal, but the neuron apoptosis was found in oppressed nerve group. The group of fake operation did not show apoptosis neurons labeled by TUNEL. The apoptosis neurons were observed after 12h in group of oppressed cauda equina nerve , and in flood tide between 24h and 48h. After 12h, the content of NO increased, with flood tide between 24h and 48h, and decreased after 72h. Conclusion:The neuron apoptosis of lumbar and sacral marrow of dogs happened early and continued long after the cauda equina never being oppressed. There is a positive correlation between the content of NO and apoptosis index.
