摘    要:对接种腹水型S180细胞的小鼠反复定量联合化疗 ,建立获得性多药耐药基因 (MDR )表达小鼠模型 ,探讨临床早期发现化疗耐药、全面定量检测耐药程度的方法。应用流式细胞术动态检测MDR表达情况及S180细胞凋亡指标 ,同法检测同期外周血淋巴细胞上述指标。结果是实验组小鼠自化疗后 1周MDR表达逐渐增高 ,自第 5周后显著性增加 (P <0 0 5 ) ;而S180细胞凋亡率、细胞杀伤率及Fas因子表达率则随化疗时间的延长而逐渐下降。MDR表达显著性增加时 ,S180细胞凋亡率等则显著性降低 (P <0 0 5 )。第 5周时实验组MDR阳性表达小鼠占 61 4 %、第 7周占 83 3 3 %。实验组外周血淋巴细胞MDR、凋亡率与S180细胞有显著相关性

Abstract:Mice injected abdominally with S180 cells were repeatedly rationed with combined chemotherapy regime in order to establish the mouse model with obtained multi drug resistance (MDR) gene expression The expression of MDR gene,apoptotic parameters and the mouse ascites S180 cell line with their peripheral blood lymphocytes were assayed by flow cytometry It was found that,in the experiment group,the expression of MDR gene gradually increased after one week of chemotherapy and markedly increased since 5 th week of treatment,but,with prolongation of treatment,the apoptotic rate,cytotoxicity of the S180 cells and the expression of Fas factor were gradually reduced At 5 th week,the percentage of mice with positive MDR gene expression was 61 4% and it was 83 3% at 7 th week. The MDR expression and apoptotic rate of peripheral blood lymphocytes were statistically correlated with those of the S180 cells
