羊源Ⅱ型胶原蛋白诱导关节炎模型相关指标的检测

目的检测羊源Ⅱ型胶原蛋白诱导关节炎模型相关指标，分析此模型的相关指标与牛Ⅱ型胶原蛋白诱导的关节炎模型的异同。方法分别用牛、羊Ⅱ型胶原蛋白免疫动物，比较两组免疫大鼠足趾容积、关节炎肿胀评分、血清细胞因子的榆测和关节滑膜病理改变。结果羊Ⅱ型胶原蛋白成功诱导出关节炎模型，与牛Ⅱ型胶原白诱导关节炎模型在造模成功率上差别无统计学意义，其中羊Ⅱ型胶原蛋白诱导组大鼠关节发病时间、关节肿胀达高峰时间及肿胀程度均早于和重于牛Ⅱ型胶原蛋白诱导组。羊Ⅱ型胶原蛋白诱导组血清细胞因子IL-1β、TNF—α升高较牛胶原蛋白诱导组更为显著（P〈0．01）。病理切片显示两模型组关节滑膜均有炎细胞浸润，滑膜细胞层次增多，排列紊乱，但牛组重于羊组。结论羊Ⅱ型胶原蛋白可诱导大鼠关节炎。

Characteristics of the Related Indicators of the Arthritis Induced by Sheep Collagen Protein

Objective To detect the relevant indicators of arthritis model induced sheep type Ⅱ collagen proteins and to analyze , analysis similarities and differences from arthritis model induced by bovine type Ⅱ, collagen. Methods Rats were immuned with type Ⅱ collagen protein extracted from cattle or sheep, the volume of raw and the degree of edema were scored and the levels of cytokines in plasma were detected. At last, the pathological changes in the synovium of joint were observed. Results The arthritis model induced by sheep type Ⅱ collagen was successfully established and the rate of success had no difference from that by bovine type Ⅱ collagen. Raws irritated by collagen protein from sheep had a quick edema but those irritated by the collagen protein from cattle had a mild one. The levels of IL - 1β and TNF - α in the plasma from the rat immuned by the collagen protein from sheep were higher than those irritated by the collagen protein from cattle. And in the pathological section, we found that the synovium of joint was infiltrated by inflammatory cells and disordered. Conclusion The sheep type Ⅱ collagen can induced arthritis in rats.
