鹅绒藤总生物碱体外抗肿瘤作用的实验研究

摘    要:目的研究鹅绒藤总生物碱(CCTA)对体外培养的肿瘤细胞的抑制作用及其可能的作用机制。方法以人宫颈癌细胞Hela、人结肠癌细胞SW480为研究对象,用不同浓度的CCTA分别对上述细胞作用24、48、72、96h后用MTr法测其抑制率,流式细胞仪测其细胞周期及凋亡率。结果CCTA对Hela细胞和SW480细胞均有不同程度的抑制作用,且呈时间和剂量依赖性;可使Hela细胞位于S期的细胞明显增多,而SW480细胞位于G0/G1期的细胞明显增多;CCTA作用后,Hela细胞和SW480细胞凋亡率均显著增加。结论CCTA可通过阻断细胞周期、诱导细胞凋亡来抑制体外培养Hda细胞和SW480细胞的生长。

Abstract:Objective To explore the etfect of CCTA in vitro on cultured tumor cells inhibition and possible mechanism. Methods Human cerical cancer hela cells、human colon cancer SW480 cells were selected as the research objects, After putting different concentrations of Cynanchum Chinense Total Alkaloid (CCTA) respectively on cells 24h ,48h ,72h, 96h. The MTT method was used to evaluate the cytotoxic effect of CCTA on the cells. Flowcytometry was used to analyze the cell cycle and apoptosis rate. Results Different concentrations of CCTA were cytotoxic to Hela cells and SW480 cells in a time - dependent and dose - dependent manner. The proportion of Hela cells in S phase was significantly increased and the proportion of SV?480 cells in Go/G1 phase was significantly increased. The FCM analysis showed that the apoptosis rate of Hela cells and SW480 cells were increased after treatment of CCTA. Conclusion CCTA can inhibit the proliferation of Hela ceils and SW480 cells in vitro, which is associated with the cell cycle and the cell apoptosis.
