摘    要:目的探讨生物可降解材料乙交酯.丙交酯共聚物(HLGA)支架与组织细胞在体内环境下的生物相容性。方法体外培养许旺细胞(SC)和神经干细胞(NSC),1周后置于PLGA支架内并移植入大鼠T9水平脊髓半横断损伤处,不同时间扫描及透射电镜下观察支架的降解及细胞的生长状况。结果扫描电镜观察髓内移植后随时间的延长,材料逐步降解。透射电镜观察材料内见再生的神经纤维、纤维细胞、纤维母细胞和巨噬细胞吞噬坏死变性的SC。结论PLGA支架与组织细胞在大鼠脊髓内具有良好的生物相容性,这为我们下一步的实验研究打下了坚实的基础。

Abstract:Objective To study the biocompatibility of PLCA with tissue cell after implantation into the injured spinal cord of rat. Methods Schwann cells and neural stem cells were harvested and cultured in vitro, Then we constructed PLGA-cell complex and implanted the complex into the injured site of T9 SCI models developed from Wistar rats. Then we observed the degradation of the porous scaffold and the growth of tissue cells in the scaffold through electron microscope at different time. Results The PLCA in the scaffold-cells complex gradually degenerated under scanning electron microscope. Regenerated axons were seen in the scaffold-cell complex. Fibrocyte fibroblast and macrophage plagocytizing necrotic Schwann cells were also visualized in the complex under transmission electron microscope. Conclusions The PLCA is a good material for central nerve tissue engineering with good biocompatibility and biodegradability. It can be used as scaffold material in tissue engineering research of SCI.
