低温抑制兔血管平滑肌细胞间缝隙连接通讯的研究

摘    要:目的探讨凝血块混合物对血管平滑肌缝隙连接通讯(GJIC)功能的影响,阐明低温治疗具有抑制此种GJIC的作用。方法培养兔血管平滑肌细胞至致密单层,分别加凝血块混合物在常温、低温下培养组,以及无凝血块混合物的常温培养组,利用划痕标记染料示踪技术(SLDT)测定GJIC水平。结果加凝血块混合物常温组(组①)染料5min扩散距离较无凝血块混合物的常温培养组(组②)远;加凝血块混合物低温组(组③)扩散距离较组①染料5min扩散距离明显缩短。①与②、①与③有统计学意义(P<0.01)。结论凝血块混合物具有增强血管平滑肌细胞GJIC的功能;低温具有降低此种GJIC功能的作用,可能是低温治疗蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)的作用机理之一。

Abstract:Objective To observe effects of sludged blood compound (SBC) on gap junctional intercellular communication (GJIC) in rabbit's vascular smooth muscle cells (VSMs) and illuminate inhibitive effects of hypothermia on this GJIC. Methods After VSMs had grown to confluence, they were divided into three groups and were exposed to different conditions. VSMs in group one were incubated in SBC and normothermic culture condition for 36 hours. VSMs in group two were exposed to normothermic culture condition for 36 hours. VSMs in group three were incubated in SBC and hypothermia culture condition for 36 hours. The effects of SBC or hypothermia on GJIC in VSMs were determined by scrape-loading and dye transfer( SLDT) technique of using a fluorescent microscope. Results In group one, the fluorescent diffusion length of VSMs was longer than group two, but, in group three, the fluorescent diffusion length of VSMs was shorter than group one. There was significant difference in fluorescent diffusion length between group one and group two (P <0.01) , between group three and group one (P <0.01). Conclusion Under the condition of this experiment, we find VSMs' GJIC was notably enhanced after applying SBC into these cultured cells. Hypothermia could inhibit this GJIC function. Such effects may be one of mechanisms of action about hypothermia in relieving CVS after SAH.
