摘    要:目的探讨进一步认识牙髓-牙本质复合体组织形态学特征的方法。方法选择20个因正畸治疗拔除的无龋前磨牙,处理后暴露牙本质,经10%福尔马林固定,8%硝酸脱钙常规组织切片。采用过碘酸—银-乌洛托品(PASM)染色套染苏木素-伊红(HE)染色,光镜下观察牙髓-牙本质复合体的组织结构。结果复染后的切片背景呈浅红色,神经纤维组织呈黑色网状结构,牙髓端有螺旋状神经纤维穿过牙本质细胞层进入前期牙本质,前期牙本质内有一定数量呈弯曲状的神经纤维分布,走行方向与牙本质小管基本一致;牙髓内的血管、细胞亦清晰显示。结论改良PASM染色结合HE复染,可获得牙髓微血管、细胞、神经组织多重结构的清晰图像。

Abstract:Objective To explore a better method to study the histological characteristics of the pulp-dentin complex. Methods 20 caries-free premolars removed for orthodontic reason were selected and the dentins were fixed by 10% formalin, decalcified by 8% nitric acid. After conventional histo-sections were stained with the Iodate-Silver-Urotropine (PASM) staining and Hematoxylin-Yihong (HE) staining, the structure of pulp-dentin complex were observed with light microscopy. Results The background of the section after double staining showed light pink and nerve tissue presented in black netlike structure, the spiral nerve fibers penetrated through the odontoblast layer and got into the pre-dentin. There were a certain number of curved nerve fibers in pre-dentin layer, distribution of which were almost the same as the tubules'. Vessels and cells in the pulp could be seen clearly. Conclusions The modified PASM staining combining with HE staining, can give a clear image of the pulp' s multiple structure, so as the microvascular, the cells and the nerve tissue.
