大鼠非麻醉状态下双尾静脉穿刺高胰岛素-正血糖钳夹实验

摘    要:目的大鼠在非麻醉状态下,采用双尾静脉穿刺术实施高胰岛素正血糖钳夹实验并与局麻下尾动静脉插管高胰岛素正血糖钳夹实验进行比较。方法普通饲料喂养和高脂高糖饲料喂养的SD大鼠各8只,喂养8周后,各分为2组,每组4只,分别进行不同的高胰岛素一正血糖钳夹实验;大鼠在非麻醉状态下,选用两条尾静脉行静脉穿刺,一条尾静脉靠近根部输入胰岛素[O.12U/(kg.h)】和葡萄糖,同时另一条尾静脉靠近尾末1/3采血,进行高胰岛素一正血糖钳夹实验。结果大鼠在非麻醉状态下双尾静脉穿刺术后血糖在正常范围内,无明显波动,无大鼠死亡;高脂高糖饲料喂养组大鼠的葡萄糖输注率显著低于对照组(P〈0.01);血糖变异系数和葡萄糖输入速率变异系数分别为5.7%和14.9%;双尾静脉穿刺术高胰岛素-正血糖钳夹实验与尾动静脉插管术高胰岛素~正血糖钳夹实验两组结果之间差异无统计学意义。结论在非麻醉状态下采用双尾静脉穿刺术行大鼠的高胰岛素正血糖钳夹实验,具有安全可行、实验结果稳定、准确、操作简便、实验周期短、术后大鼠恢复快、可多次重复进行等优点;大鼠饲喂高脂高糖饲料可诱发明显的胰岛素抵抗。

Hyperinsulinemic-euglycemic clamp adopting venopuncture of double venae caudalis in rats without anesthesia

Abstract:Objective To compare hyperinsulinemic-euglycemic clamp adopting venopuncture of double venae caudalis in rats without anesthesia with that of adopting catheterization of arteria caudilis and vena caudalis in rats under local anesthesia. Methods 16 SD rats were divided into 4 groups and each group contained 4 rats. 2 groups were fed with standard diet and the other 2 groups were fed with high-fat and high-sucrose diet. Eight weeks later, one vena caudalis of rats without anesthesia was used to inject insulin [0.12 U/(kg.h)] and glucose and the other vena cau- dalis was used to obtain blood samples for performing hyperinsulinemic-euglycemic clamp respec- tively. Results After the venopuncture of vena candalis, the blood glucose of the rats was within the normal range without any significant fluctuation, and no rat died. The glucose infusion rate (GIR) of the high-fat and high-sucrose diet group was significantly lower than that of the normal diet group (P 〈0.01). The blood glucose coefficient of variation and GIR coefficient of variation were 5.7% and 14.9% respectively. There was no significant difference between the two techniques. Conclusion Compared with hyperinsulinemic-euglycemic clamp adopting catheterization of ar- teria caudilis and 'vena caudalis in rats under local anesthesia, hyperinsulinemic-euglycemic clamp adopting venopuncture of double venae caudalis in rats without anesthesia is safer, more feasible, easier to perform, and has shorter experimental period and lower mortality. Rats fed with high-fat and high-sucrose diet are susceptible to insulin resistance.
