阿魏酸钠对抗谷氨酸诱导的鼠皮层神经元凋亡作用的影响

摘    要:目的探讨MEK/ERK信号转导通路是否参与了阿魏酸钠(SF)对抗谷氨酸诱导的鼠皮层神经元凋亡作用。方法以谷氨酸诱导大鼠皮层神经元凋亡为模型。采用Westernblot观察Bcl-2、caspase-3、磷酸化ERK1/2表达的改变。结果阿魏酸钠能够显著降低谷氨酸诱导的神经细胞caspase-3的表达,提高Bcl-2、磷酸化ERK1/2的表达。PD98059可以减弱SF的保护作用。结论 MEK/ERK1/2通路参与阿魏酸钠对抗谷氨酸诱导的鼠皮层神经元凋亡。

Effect of sodium ferulate inhibits glutamate-induced rat cortical neurons apoptosis

Abstract:Objective To investigate whether MEK/ERK signal pathway be involved in anti-apoptotic effect produced by sodium ferulate. Methods Apoptosis was induced by glutamate in cultured rat cortical neurons. Bcl-2,caspase-3 and phos- pho-ERK1/2 were detected by Western blot technique. Results Sodium ferulate significantly inhibited the glutamate-induced downregulation of phospho-ERK1/2 and Bcl-2 expression in cultured cortical neurons. At the same time reduced the glutamateinduced caspase-3 expression. PD98059 reduced the protective effect of sodium ferulate. Conclusion Sodium ferulate protect cortical neurons from glutamate-induced apoptosis. MEK/ERK signal pathway is contributed to the effect.
