山东不同产地北虫草中麦角甾醇提取工艺与含量测定的研究

摘    要:目的:建立北虫草子实体质量评价的HPLC方法,研究和比较山东区域不同产地的北虫草子实体中麦角甾醇的提取工艺和含量。方法:采用正交试验设计筛选北虫草中麦角甾醇最佳提取工艺,利用HPLC法定量分析10个不同产地北虫草子实体中麦角甾醇的含量;采用色谱柱为Promosil-C18柱,流动相为甲醇,流速为1.0 mL/min,柱温为30℃,检测波长为282 nm。结果:北虫草中麦角甾醇的最佳提取最佳工艺为A2B3C2,即料-液比为1∶40,40℃时超声提取75 min;不同产地的北虫草中麦角甾醇含量范围为0.25%~0.68%。结论:本方法操作简便,结果准确可靠,可用于北虫草子实体中麦角甾醇的含量测定,为北虫草的质量评价提供了参考。

Studies on Extraction Process and Assaying of Ergosterol in Cordvceps Militaris from Different Habitats in Shandong

Abstract:Objective: To establish a HPLC method to evaluate the quality of the fruiting body of Cordyceps militaris, and to study the extraction process of ergosterol and comparison from different habitats of Shandong area in Cordyceps militaris and content. Methods: Using the orthogonal design to select the best extraction process of ergosterol in Cordyceps militaris, analyse the ergosterol content in Cordyceps militaris from 10 different habitats by HPLC statutory amount; using Promosil-C18 column, the mobile phase of methanol, the flow rate was 1 mL/min, the column temperature was 30 ℃ and the detection wavelength was 282 nm. Results: The optimum extracting ergosterol in Cordyceps militaris is A2B3C2, namely the material liquid ratio 1:40, 75 min at 40 ℃ ultrasonic extraction; content of ergosterol in Cordyceps militaris from different habitats were between 0.25% ang 0.68%. Conclusion: The method is simple, accurate and reliable, and can be used for the determination of ergosterol in Cordyceps militaris, providing the reference for the quality evaluation of Cordyceps militaris.
