黄芪联合丹参注射液对大鼠脑出血半暗带区神经元凋亡抑制作用的研究

摘    要:目的 探讨黄芪联合丹参注射液对大鼠脑出血半暗带区神经元凋亡的抑制作用.方法 应用脑立体定位仪于大鼠尾壳核注入肝素化Ⅶ型胶原酶建立脑出血模型.将大鼠随机分为正常组、出血对照组和治疗组.分别对各组大鼠进行神经行为学、吉姆萨(Giemsa)染色、免疫组化染色及细胞超微结构等实验观察.结果 各治疗组大鼠神经行为学评分较出血对照组明显减小(P<0.05).各治疗组脑出血灶周围细胞凋亡数量及程度较出血对照组明显降低.与相应出血对照组比较,各治疗组Bcl-2阳性表达均明显升高(P<0.05),Bax及Caspase-3阳性表达均明显降低(P<0.05);与相应术后24 h给药治疗组比较,各术后6h给药治疗组Bcl-2阳性表达均明显升高(P<0.05),Bax及Caspase-3阳性表达均明显降低(P<0.05);与相应3d各时间给药治疗组相比,7d给药治疗组Bcl-2阳性表达均明显升高(P<0.05),Bax及Caspase-3阳性表达均明显降低(P<0.05).各治疗组与出血对照组比较,神经元细胞线粒体、核膜、内质网等结构相对完整.结论 黄芪联合丹参注射液对大鼠脑出血半暗带区神经元凋亡具有较好的抑制作用.

Abstract:Objective To study the inhibition of Astragalus and Salviae miltiorrhizae injection on the neuronal apoptosis of the penumbra around the Intracerebral Hemorrhage(ICH) in rats.Methods Collagenase was injected into caudate nucleus to induce rat model of ICH with brain stereotaxic instrument.Rats were randomly divided into normal group,ICH control group and treatment group.Neurological behavior,giemsa staining,immunohistochemical staining and cell ultrastructure were observed.Results Neuronal behavior score in treatment group rats was much less than that of ICH control group(P<0.05).Compared with the ICH control group,the number and extent of the cell apoptosis around the intracerebral hemorrhage of the treatment group was significantly reduced.Compared with the corresponding ICH control group,the expression of the Bcl-2 in each treatment group was significantly increased(P<0.05),Bax and Caspase-3 expression were significantly reduced(P<0.05);compared with the corresponding 24 h after surgery drug treatment group,the expression of the Bcl-2 of each 6h after surgery drug treatment group was significantly increased(P<0.05),Bax and Caspase-3 expression were significantly reduced(P<0.05);compared with the 3 d drug treatment group,the expression of the Bcl-2 in each 7d drug treatment group were significantly increased(P<0.05),Bax and Caspase-3 expression were significantly reduced(P<0.05).Compared with the corresponding ICH control group,the injury of neuronal mitochondria,nuclear membrane,endoplasmic reticulum and other structures in each treatment group was significantly alleviateal relatively intact.Conclusion There is a good inhibition on the neuronal apoptosis of the penumbra around the intracerebral hemorrhage in rats for the Astragalus and Salviae miltiorrhizae injection.
