云南小耳猪胰岛细胞的分离和纯化方法

目的探索云南小耳猪胰岛细胞分离和纯化的有效方法.方法采用胶原酶胰导管注射负荷技术,静止消化与物理消化相结合消化分离小耳猪胰腺;不连续密度梯度比重离心纯化,胰岛素释放试验检测胰岛细胞功能.结果胰岛收获量为（4580±257）IEQ/g胰腺,纯化后为（3660±449）IEQ/g胰腺,纯度为80%左右,胰岛素释放反应良好.结论采用本方法消化分离小耳猪胰岛,腱岛细胞产量、纯度较高,功能良好,为进一步成人胰岛细胞的分离纯化提供了实验依据.

Pancreatic Islet Isolation and Purification of Yunnan Miniature Pig

Objective To explore an effective way of pancreatic islet cell isolation and purification of Yunnan Miniature Pig.Methods Islets were isolated using digestion by pancreatic duct injection of collagenase,and still digestion combined with the physical digestion.Islet cells were purified by the proportion of discontinuous density gradient centrifugation;it＇s function was detected by insulin release test.Results（4 580±257）IEQ per gram of pancreas were obtained,after purification the recover were（3 660±449）IEQ per gram of pancreas;the purify of the final preparation was about 80%.The release of insulin was well received.Conclusions The production and purity of island cells were high and well-functioning by this method.It provides experimental evidence for the isolation and purification of adult islet cells.
