IBS中药复方不同方法制备样品的比格犬灌胃给药吸收入血成分定性差异及其意义

摘    要:目的:通过对同一种复方中药采用水煎和乙醇提取两种不同方法制备样品进行血中吸收成分的比较,为目前中药药代动力学研究所面临的技术难点提供解决方案。方法:采用液一液萃取净化、996-二极管阵列检测高效液相色谱法测定血浆样品。结果:以多种剂量灌胃给予比格犬IBS复方水煎剂,血浆样品中均未检测到任何吸收入血的成分,但是用同样的方法给予比格犬IBS复方乙醇提取样品,在剂量较水提样品低约27倍时,却至少可以检测到20种化学成分。结论:比格犬灌胃给予IBS中药复方乙醇提取样品和给予水提取样品相比,两者吸收入血成分具有重大差异,这一差异对揭示目前中药复方药代动力学研究难点以及提高中药的临床疗效可能潜在重要意义。

Abstract:Authors qualitatively analyzed the variations of medicinal ingredients in beagle dog's plasma, after given orally the products made from the same formula, but prepared in a different way. In the study, Beagle dog's plasma samples were analyzed, after treated by liquid extraction in a Waters HPLC system equipped with the PDA detector. The mobile phase was either made up of methanol water, or acetonitrile and water. The retention time of HPLC, and UV spectrum were used the qualitative criteria for judging the ingredients in the blood samples. It is found that no interesting ingredients were detected in the plasma samples after given with the IBS formula treated with water at different dosages. However, at least 20 compounds could be detected in the blood samples in the group treated with IBS formula extracted with ethanol at a dosage 27 times lower, compared with the water treated IBS formula group. Authors believe that plasma ingredients can be significantly varied if treated with the water extracted IBS formula, or with the ethanol extracted IBS formula. The phenomenon may explain the insufficient confidence in telling the effectiveness of TCM drugs.
