新型骨基质材料复合骨髓基质干细胞体外培养的形态学观察

摘    要:目的:新型有机-无机复合类骨基质材料(New Bone-matrix Material,NBM)的研制及其细胞相容性的初步检测,为骨组织工程探寻一种新型复合类细胞外基质材料提供实验依据。方法:应用组织培养技术,对兔骨髓基质干细胞(Bone Marrow Stromal Cell,BMSC)和NBM体外联合培养进行相差显微镜观察和扫描电镜观察。结果:骨髓基质干细胞可以在NBM材料上发生良好的粘附、增殖,并能长入材料的孔隙内,分泌大量的细胞外基质成份。结论:NBM是一种新研制的有机-无饥复合类骨基质材料,具有网状孔隙结构;在体外与BMSC复合培养时表现出良好的细胞相容性,对细胞的粘附、增殖和功能表达未见明显影响。

Abstract:Objective: A compound new bone matrix material (NBM) composed of both organic and inorganic materials was designed for bone tissue engineering, and the biocompatibility of NBM with bone marrow stromal cells (BMSC) was investigated. Methods: The morphological characteristics of BMSC cultured with NBM in vitro was observed using phase contrast microscope and scanning electron microscope. Results: BMSC could attach and proliferate well on the NBM, secrecting lots of extracellular matrix. Conclusion: NBM is a new compound bone matrix material constituted of organic and inorganic materials, which pocesses a morphological structure of three dimensional foam. NBM showed a good biocompatibility with BMSC in vitro. There was no signs of inhibition of NBM material to the attachment, proliferation and functional expression of BMSC.
