摘    要:目的:对脑内兴奋性氨基酸的变化进行定位研究。方法:应用Beckman-6300氨基酸分析仪和601黄金系统色谱工作站,对梭曼惊厥后不同时相大鼠的新鲜脑组织进行定位检测。结果:梭曼诱发惊厥后脑内谷氨酸和天门冬氨酸的水平显著降低,谷氨酸下降最明显的是惊厥30min后的大脑皮质和海马,分别是正常组的53.2%和52.6%,小脑的嗅球内谷氨酸水平显著下降,天门冬氨酸更易受梭曼中毒的影响,惊厥后5、30、9

Abstract:Objective: To detect changes in levels of glutamate and asparate in rat discrete brain regions which may be involved in the mechanism of seizure initiation and maintenance in soman induced seizures. Methods: Total amino acids in freshly prepared brain regions in rats at different time phases after seizure dissected under microscope were detected by using a Beckman 6300 analyzer and 601 Gold system for spectrochrome analyzer. Results: The levels of total glutamate and asparate were significantly decreased in the rat brain after soman induced seizures. The lowest level of glutamate was observed in cerebral cortex and hippocampus in 30 min after seizure initiation. It decreased in other regions including cerebellar cortex and olfactory bulb. Asparate appeared to be more vulnerable to soman intoxication. The levels of asparate in 6 brain regions including striatum cerebral cortex, hippocampus, midbrain, cerebellum, diencephalon and olfactory bulb at 5, 30 and 90 min after seizure were significantly decreased as compared with the control. Conclusion: The excitatory amino acid transmitter activity may play an important role in soman induced seizures.
