化毒丹对小鼠迟发型超敏反应的影响

摘    要:目的:研究化毒丹对小鼠迟发型超敏反应(DTH)的影响。方法:50只Balb/c鼠随机分为正常对照组、DTH模型组及化毒丹高剂量组(1.0mg/g)、中剂量组(0.5mg/g)和低剂量组(0.25mg/g)。除空白组小鼠的左侧足垫注射生理盐水0.05ml外,其余各组小鼠左侧足垫注射10%绵羊红细胞的生理盐水悬液0.05ml,同时,对化毒丹高、中、低剂量组小鼠给予0.5ml化毒丹灌胃,连续灌胃5d后,用0.05ml 10%SRBC.NS攻击小鼠右侧足垫(空白组注射0.5ml生理盐水),24h后测量小鼠足垫肿胀程度。采用ELISA法检测血清中肿瘤坏死因子(TNF)水平,采用噻唑兰(MTT)法检测淋巴细胞的活性。结果:与正常对照组比较,DTH模型组及化毒丹高、中、低剂量组小鼠足垫肿胀程度明显增加(P〈0.05~0.01),与DTH模型组相比较,化毒丹各剂量组小鼠足垫肿胀程度明显增加(P〈0.001);与正常对照组比较,DTH模型组、化毒丹高剂量组小鼠血清中TNF水平明显降低(P〈0.05~0.01),与DTH模型组相比较,化毒丹中、低剂量组小鼠血清中TNF水平明显增加(P〈0.001);与正常对照组比较,化毒丹中剂量组小鼠脾脏淋巴细胞活性明显降低(P〈0.05),化毒丹低剂量组小鼠脾脏淋巴细胞活性明显增加(P〈0.05)。结论:化毒丹对小鼠DTH具有双向调节作用。

Abstract:Objective: To observe the effects of HUA Dudan against the delayed-type hypersensitivity in mice.Methods: The delayed-type hypersensitivity animal model in mice was established by injection of sheep red blood cell(SRBC).We tested the level of tumor necrosis factor(TNF) in serum and the activity of lymphocyte and observe the level of swell of foot in mice.Results: Contrasted to normal group,each dose of Hua Dudan can increase the degree of swell as well as model group(P<0.05~0.01) and the high dose of Hua dudan can decrease the lever of TNF in serum(P<0.05~0.01),the middle-dose of Hua Dudan may decrease the activity of lymphocyte(P<0.05);Contrasted to model group each dose of Hua Dudan can increase the degree of swell(P<0.001) and middle and low dose of Hua dudan can increase the level of TNF in serum(P<0.001),low dose of Hua dudan can enhance the activity of lymphocyte(P<(0.05)).Conclusions: HUA Dudan has the function of bi-regulate the delayed-type hypersensitivity in mice.
