凋亡抑制蛋白Livin在肾癌组织中的表达及其意义

目的观察凋亡抑制蛋白Livin在肾癌组织内的表达情况,分析Livin的表达与肾癌临床病理特征之间的关系。方法应用免疫组化法和Westernblot技术观察Livin在肾癌组织内的表达。结果 Livin在肾癌组织及正常肾组织中均有表达,Livin蛋白表达于肿瘤细胞胞质,在48例肾癌组织中Livin阳性表达率为79.2%,对照组16例正常肾组织中Livin阳性表达率为18.8%,二者之间差别具有统计学意义(<0.05)。Westernblot检测结果表明在肾癌组织中Livin蛋白的表达量显著高于正常肾组织的表达水平(<0.05)。Livin表达与患者性别、年龄、肿瘤分化程度无关,与临床分期和淋巴结转移显著相关(<0.05)。Kaplan-Meier生存分析结果表明,Livin阴性患者无瘤生存率明显高于Livin阳性患者(<0.05)。结论 Livin在肾癌组织高表达与肾癌的进展和淋巴结转移密切关系,可能预示肾癌复发。

Expression and significance of apoptosis protein inhibitor-livin in renal cell carcinoma

Objective To investigate the relationship between livin expression and clinicopathological characteristics in renal carcinoma we observed the expression of livin in renal carcinoma.Methods The expression of livin in renal carcinoma was detected by immunohistochemical staining and Western blot.Results Livin protein expression was observed in renal carcinoma and normal renal tissue,but livin expressed predominantly in the cytoplasm of tumor cells,the positive expression rate was 79.2% and 18.8% in 48 renal carcinoma and 16 normal renal tissue respectively with obvious differnces between them(<0.05).The results of Western blot showed that the expression level of livin protein in renal carcinoma was higher than that in normal renal tissue(<0.05),with significant correlation between livin expression and clinical stage and lymph node metastasis(<0.05),but with no relationship of livin expression with sex,age and tumor differentiation grade.Kaplan-Meier analyses indicated that relapse-free survival rate was higher in livin negative expression patients than in positive expression patients(<0.05).Conclusion High expression of livin in renal carcinoma is closely correlated with tumor progress and lymph node metastasis,examination of livin protein might be beneficial for diagnosis of relapse of renal carcinoma.
