摘    要:目的探讨重组人促红细胞生成素(recombinant human erythropoietin,rHuEPO)对脑缺血再灌注小鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法 96只昆明小鼠随机分为3组,每组32只:假手术组(Sham);rHuEPO治疗组;缺血再灌注组(I/R)。每组依据缺血后再灌注不同时间点再分6h、24h、3d、7d五个小组。免疫组化和Westernblot检测各组小鼠VEGF的表达,TUNEL法检测海马细胞凋亡情况。结果缺血再灌注组海马VEGF蛋白表达量和凋亡细胞数明显高于假手术组(<0.05),与缺血再灌注组相比,rHuEPO治疗组VEGF蛋白表达量明显升高(<0.05),凋亡细胞数则明显少于缺血再灌注组(

Abstract:Objective To study the expression of vascular endothelial growth factor(VEGF) in hippocampus of cerebral ischemia-reperfusion mice after recombinant human erythropoietin(rHuEPO)was injected.Methods 96 Kunming mice were randomly divided into normal control group,ischemia-reperfusion group,recombinant human erythropoietin group at different time points(6h,24h,3d and 7d)on average.The expression of VEGF in hippocampus was examined by immunochemistry and western blot methods,the apoptotic hippocampus cells were stained by TUNEL.Results Both the expression level of VEGF and the number of apoptosis cells increased significantly in I/R group than in sham group( 0.05),however,the expression level of VEGF was higher and the number of apoptosis cells was lower in recombinant human erythropoietin group than in I/R group(0.05).Conclusion The repairment of recombinant human erythropoietin on the cerebral ischemia-reperfusion injury might be related to the upexpression of VEGF.
