核转录因子κB在非小细胞肺癌中的表达及意义

目的研究核转录因子κB（nucleus factor-кB,NF-кB）P65亚单位在非小细胞肺癌（non-small cell lung cancer,NSCLC）中的表达及与非小细胞肺癌临床病理特征的关系及意义。方法采用免疫组织化学染色方法（PV-6000通用型二步法）检测62例NSCLC组织和20例癌旁肺组织中NF-кBP65蛋白。结果P65蛋白在NSCLC组织中阳性表达率为79%（49/62）,明显高于癌旁肺组织50%（10/20）,差异有统计学意义（P〈0.05）。P65蛋白与NCSCLC细胞分化程度、肺癌分期和淋巴结转移有关,差异有统计学意义（P〈0.05）。结论NF-кBP65蛋白高表达与NSCLC的发生、细胞增殖及浸润转移相关,可作为预后观测的指标和分子治疗的新靶点。

Expression and Clinical Significance of nucleus factor-kB P65 Protein in Non-small Cell Lung Cancer

Objective The aims of this study were to evaluate expression levels of nucleus factor-кB P65 Protein in non-small cell lung cancer （NSCLC） and to elucidate its clinical significance and prognostic value for patients with NSCLC. Methods Using 62 tumor tissue specimens and 20 noncancerous tissue specimens from 62 patients with NSCLC who underwent surgery,we investigated the expression of nucleus factor-кB P65 Protein by immunohistochemical methods.Slides were scored semi-quantitatively based on staining intensity and distribution,and results were compared with clinical pathologic parameters. Results Forty-nine（79%） of 62 tumors had positive nucleus factor-кB P65 Protein expression.Ten（50%） of 20 noncancerous tissue had positive nucleus factor-кB P65 Protein expression.There was significant difference in the expression of nucleus factor-кB P65 Protein between tumor tissues and noncancerous tissues（P〈0.05）. Higher expression levels of nucleus factor-кB P65 were associated with tomor poorer differentiation,higher clinical stsge and lymph node metastasis. Conclusion Higher expression levels of nucleus factor-кB P65 was closely correlated with oncogenesis,proliferation,invasion and metastasis.
