小鼠动脉粥样硬化斑块中RGS5等基因的表达改变及黄芩素干预的实验研究

摘    要:目的观察黄芩素对小鼠动脉粥样硬化(AS)模型斑块内RGS5等基因表达的影响。方法选用APOE-/-小鼠30只,体重18~22 g,随机分别对照组、模型组和干预组,每组10只,对照组给予APOE-/-普通饮食,模型组以高脂饲料喂养,干预组在高脂饮食中添加黄芩素(0.5%)。造模给药12周后,检测血脂,用Real Time-PCR方法检测对照组、模型组和用药组主动脉壁粥样硬化斑块中RGS5、PPARδ、MCP-1、NF-κB mRNA的表达。结果模型组血脂升高,斑块中RGS5 mRNA表达下调,PPARδ、MCP-1、NF-κB mRNA表达升高(P0.01)。黄芩素降低了高脂血症并逆转了上述基因表达,延缓了AS进程,差异均具有统计学意义(P0.05)。结论 RGS5等基因在AS的进程中具有重要的调控作用,黄芩素通过降低RGS5的表达,延缓AS的发生。

Abstract:Objective]To study the effect of baicalein on the expression of RGS5 mRNA in atherosclerosis(AS) plaque of mice.Methods]A total of 30 APDE-/-Mice 18-22 g were randomly divided into three groups(10 in each group) as follows: the control group fed with APOE-/-normal diet;the model group fed with high-fat diet;the treatment group fed with high-fat diet and 0.5% baicalein.After 12 weeks,the levels of serum lipid and the RGS5,PPARδ,MCP-1,NF-κB mRNA expression were detected by Real Time-PCR.Results] An increase of lipid was detected and the expressions of the genes were also obvious changed in model group(RGS5 was down regulated;PPARδ,MCP-1,NF-κB were up regulated;P<0.05).Baicalein reduced blood lipid,reversed the expression of above genes,and delayed the AS process(P<0.05).Conclusion]RGS5 gene plays an important regulatory role in the process of AS.Baicalein can delay occurrence of AS by reducing the expression of RGS5.
