基质窖蛋白-1在食管鳞癌中的表达及与自噬的关系

摘    要:目的 探讨基质窖蛋白-1(caveolin-1,Cav-1)在人食管鳞癌组织中的表达以及与自噬标志物——微管相关蛋白轻链3B(microtubule associated protein light chain 3B,MAP-LC3B,简称LC3B)的关系及其临床病理意义.方法 利用量子点免疫荧光组织化学技术检测基质Cav-1和LC3B蛋白在76例食管鳞癌组织和15例非癌变食管黏膜上皮组织中的表达.结果 基质Cav-1和LC3B蛋白在食管鳞癌组织的阳性表达率分别为28.9%和64.5%,与非癌变食管黏膜上皮组织比较,差异均有统计学意义(P<0.05).基质Cav-1的阳性表达与食管鳞癌TNM分期和淋巴结转移均显著相关(P<0.05),而与其他临床病理参数均无关(P>0.05).LC3B与肿瘤浸润范围、TNM分期和淋巴结转移均显著相关(P<0.05).基质Cav-1和LC3B蛋白在食管鳞癌中的表达呈正相关(r=0.353,P=0.002),二者缺失表达能更好地预示淋巴结的转移.结论 基质Cav-1和LC3B表达缺失可协同促进食管鳞癌的形成和恶性进展.

Expression of stromal caveolin-1 in esophageal squamous cell carcinoma and its correlation with autophagy

Abstract:Objective To investigate stromal caveolin-1 (Cav-1) protein expression and its correlation with autophagy marker-microtubule associated protein light chain 3 ( LC3B ) in human esophageal squamous eel1 carcinoma ( SCC ) and its clinicopathologic significance. Methods Quantum dots immunofluorescent histochemistry was used to detect the expression of stromal Cav-1 and LC3B in 76 cases of esophageal SCC and 15 samples of noncancerous esophageal mucous epithelium tissues. Results The positive rates of stromal Car-1 and LC3B proteins in esophageal SCC were both significantly lower than those in noncancerous esophageal mucous epithelium tissues ( 28.9% vs 100.0% , 64. 5% vs 100. 0% ,both P 〈 0. 05 ). Stromal Cav-1 protein expression was siginificantly correlated with TNM stage and lymph node metastasis (P 〈0.05), but not correlated with other clinicopathologic parameters (P 〉 O. 05). LC3B protein level was associated with tumor invasive depth, clinical TNM stage and lymph node metastasis ( P 〈 0.05 ). There was a positive correlation between stromal Cav-1 and LC3B in esophageal SCC tissues ( r = 0. 353, P 〈 0.05 ). Conclusion Absent expression of stromal Cav-1 and LC3B proteins may synergistically promote the development and malignant progression of esophageal squamous cell carcinoma.
