神经节苷脂在大鼠脑外伤中对GAP-43蛋白表达的影响

摘    要:目的:研究神经节苷脂对大鼠自由落体脑损伤后脑内生长相关蛋白(GAP-43)表达的影响.方法:将S-D雄性大鼠随机分为4组:即正常对照组,假手术组,脑损伤(TBI)模型组,TBI后GM-1治疗组.复制大鼠自由落体脑损伤模型,在外伤后1、3、7、14 d,利用HE染色观察神经细胞形态和免疫组化染色检测神经细胞内GAP-43蛋白表达的变化.结果:脑内GAP-43蛋白的表达水平在头部损伤后逐渐升高,以伤后第三天、第七天最明显,随后有所下降,至第十四天仍轻度增高,TBI组各时段的GAP-43阳性蛋白表达均高于同时段正常对照组及假手术组,差异有统计学意义;神经节苷脂治疗组能促进TBI后GAP-43蛋白的表达,与同时段模型组对比差异有统计学意义,P均<0.05.结论:GAP-43蛋白在大鼠受到脑外伤后表达增高,可促进中枢神经损伤后的再生.神经节苷脂GM-1在TBI后可促进脑内GAP-43的表达,有稳定细胞膜、减轻细胞水肿、促进神经再生的作用.
