HLA-G在无症状沙眼衣原体和解脲支原体感染患者早孕绒毛滋养细胞的表达及意义

摘    要:目的:研究无症状沙眼衣原体和解脲支原体感染患者HLA-G在母胎界面的表达变化,及其对早孕绒毛滋养细胞侵蚀力的影响。方法:采用SP法对正常早孕和无症状沙眼衣原体及解脲支原体感染患者早孕绒毛组织中HLA-G免疫组化染色并观察HLA-G的定位和分布;采用免疫印迹法检测正常与无症状沙眼衣原体和解脲支原体感染患者早孕绒毛组织中HLA-G的表达情况;分离正常与无症状沙眼衣原体和解脲支原体感染患者绒毛外滋养细胞并进行培养和鉴定,应用免疫细胞化学法检测HLA-G的表达。结果:免疫组化结果显示正常妊娠时HLA-G主要表达于绒毛滋养细胞;免疫印迹灰度分析显示,对比正常妊娠无症状沙眼衣原体和解脲支原体绒毛感染者的早孕绒毛中HLA-G蛋白表达减弱;培养的正常早孕绒毛外细胞滋养细胞中HLA-G呈强阳性表达,而无症状沙眼衣原体和解脲支原体感染者其早孕绒毛外细胞滋养细胞中HLA-G表达减弱。结论:无症状沙眼衣原体和解脲支原体感染能对早孕绒毛HLA-G表达产生影响,造成母胎界面局面免疫功能紊乱,滋养细胞容易受到母体的攻击,从而影响到滋养细胞的侵蚀能力。

Abstract:Objective:To investigate the expression of HLA-G in the villus and extra-villous trophoblasts in patients of asymptomatic infection of CT and Uu.Methods:Immunohistochemistry was used to detect the HLA-G to show the distribution.The expressions of HLA-G protein were studied by Western blot.Highly purified extra-villous trophoblasts were detected for HLA-G in vitro by immunocytochemistry in patients of asymptomatic infection of CT and Uu.Results:In normal pregnancy HLA-G was strongly expressed in the villous tissue while they were weaker in the villus of asymptomatic infection of CT and Uu(P<0.01).HLA-G was found expressed in extra-villous trophoblasts in both normal pregnancies and those with asymptomatic infection of CT and Uu.In normal pregnancy HLA-G was strongly expressed in the extra-villous trophoblasts while they were weaker in those of asymptomatic infection of CT and Uu(P<0.01).Conclusion:The expression of immune-related molecules like HLA-G in the villus may mediate and contribute to invasion of extra-villous trophoblasts which take an important role in pregnancy while asymptomatic infection of CT and Uu may affect the invasive function of extra-villous trophoblasts.
