黑大蒜提取物对衰老小鼠抗氧化活性及免疫功能的影响

摘    要:目的 探讨黑大蒜提取物对衰老模型小鼠抗氧化活性超氧化物歧化酶(SOD)和丙二醛(MDA)以及免疫功能的影响.方法 30只BALB/c小鼠随机分成三组,即生理盐水对照组,D-半乳糖诱导衰老组,黑大蒜提取物组,每组10只.除正常对照组外,其余两组诱导衰老模型.各组灌胃给药,正常对照组及衰老模型组灌服生理盐水,黑大蒜提取物组灌服黑大蒜提取物,6周后检测血清、肝和脑组织的SOD和MDA,并且检测NK细胞杀伤活性、脾细胞NO分泌水平、脾细胞培养上清细胞因子分泌水平.结果 黑大蒜提取物组与诱导衰老组相比,对衰老小鼠血清和肝的SOD含量有明显上调功能(P=0.0471,P=0.0315),但对脑的SOD影响不明显,差异无统计学意义(P=0.0523);对衰老小鼠血清、脑和肝的MDA含量有明显下调功能(P=0.0262,P=0.0461,P=0.0360).黑大蒜提取物组与诱导衰老组相比,NO分泌水平和NK细胞杀伤活性明显升高(P=0.0117,P=0.0038);Th1型细胞因子(IL-2、IFN-γ和TNF-α)分泌水平明显升高(P=0.0042,P=0.0012,P=0.0024);Th2型细胞因子分泌水平明显降低(P=0.0481).结论 黑大蒜提取物可明显提高衰老模型小鼠抗氧化活性,增强NK细胞和巨噬细胞活性,诱导Th1型免疫应答.

Effects of black garlic extracts on the anti-oxidant activity and immune function in aged mice

Abstract:Objective To investigate the effects of black garlic extracts on the anti-oxidant activity [ superoxide dismutase (SOD) and malondialdehyde (MDA)] and immune function in aged mice induced by D-galactose. Methods BALB/c mice were randomly divided into NS control group, aged group, and black garlic extract group, with 10 mice per group. Besides NS control group, other groups were induced by D-galactose. Mice were orally administrated with black garlic extracts, and after 6 weeks of ad- ministration, the activities of NK ceils, the NO levels in splenocyte, the cytokine levels in splenocyte, the levels of SOD and MDA in serum, liver and brain were detected. Results SOD in serum, and liver in the black garlic extract group was significantly higher than that in the aged group, respectively ( P =0. 0471, P =0. 0315), but brain SOD was no significant difference between two groups ( P = 0. 0523 ). MDA in serum, liver, and brain in black garlic extract group was significantly lower than that in the aged group, respectively ( P = 0. 0262, P = 0. 0461, P = 0. 0360). The activities of NK cells and the NO levels in splenocyte were significantly higher than those in the aged group ( P =0. 0117, P =0. 0038). The secretion levels of TNF-α, IFN-γ, and IL-2 in splenocyte were significantly higher than those in the aged group ( P =0. 0042, P =0. 0012, P =0. 0024), but the secretion levels of IL-4 in splenocyte was lower than that in the aged group ( P = 0. 0481 ). Conclusions The black garlic extracts had a certain stimulating effects on Th1 type immune responses, and enhanced anti-oxidant activity in the aged mice induced by D-galactose.
