针刺对2型糖尿病肥胖大鼠瘦素、胰岛素抵抗和脂联素水平影响的实验研究

摘    要:目的:探讨针刺对2型糖尿病肥胖大鼠瘦素、胰岛素抵抗和脂联素水平影响的作用机制。方法:采用2型糖尿病肥胖大鼠模型,分为正常对照组、模型对照组、针刺组和盐酸西布曲明组4组,针刺组给予针刺中脘、天枢、足三里和三阴交,盐酸西布曲明组给予盐酸西布曲明按3mg/kg灌胃,正常对照组继续常规饲料饲养,不做处理,模型对照组按10mL/kg生理盐水灌胃。结果:干预前Lee’s指数模型对照组、针刺组、盐酸西布曲明组与正常对照组比较,差异均有显著性意义(P<0.05)。干预后,针刺组、盐酸西布曲明组Lee’s指数、脂肪湿重、脂肪系数均低于模型对照组(P<0.05)。而模型对照组、针刺组、盐酸西布曲明组的脂肪湿重、脂肪系数仍然高于正常对照组(P<0.05)。针刺后针刺组总胆固醇(TC)、甘油三酯(TG)、胰岛素、瘦素水平显著下降,脂联素水平上升,与模型对照组比较,差异有显著性或非常显著性意义(P<0.05,P<0.01),但仍达不到正常大鼠的水平。针刺组与盐酸西布曲明组之间各指标无显著差异。结论:针刺治疗可显著降低2型糖尿病肥胖大鼠的瘦素水平,升高脂联素水平,并改善其他肥胖指标。

Influence of Acupuncture on Leptin, Insulin-resistance and Adiponectin of Type 2 Diabetic Obesity Rats

Abstract:Objective. To investigate the mechanism of acupuncture in affecting leptin, insulin resis- tance and adiponectin levels of type 2 diabetic obesity rats. Methods= Type 2 diabetic obesity rat model was adopted. Rats were divided into four groups, normal control group, model group, acupuncture group and sibutramine hydrochloride group. Acupuncture group was acupunctured in acupoints of Zhongwan, Tianshu, Zusanli and Sanyinjiao. Sibutramine hydrochloride group was given gastric gavage of sibutramine hydrochloride by 3 mg/kg. Normal control group was still fed with conventional forage, and had no medi- cation. Model group was given normal saline orally by 10 mg/kg. Results= Before treatment, Lee's index in model group, acupuncture group, and sibutramine hydrochloride group had no statistically significant difference from that in the normal control group(P 〈 0.05). After treatment, Lee's index, fat wet weight, fat coefficient in the acupuncture group and sibutramine hydrochloride group were lower than those in the model group(P 〈 0.05). In model group, acupuncture group and sibutramine hydrochloride group, fat wet weight and fat coefficient were still higher than those in the normal control group(P 〈 0.05). In acupuncture acupuncture group, TC, TG, insulin and leptin levels were significantly decreased, adiponectin level was increased as compared with the model group(P 〈 0.05, P 〈 0.01), but their levels were still higher or lower than those of normal rats. There were group and sibutramine hydrochloride group. level, increase adiponectin level and improve ty rats. no significant differences of indicators between acupuncture Conclusion, Acupuncture can significantly decrease leptin other obesity indexes of experimental type 2 diabetic obesity rats.
