己酮可可碱对血管内皮细胞炎性反应的影响

摘    要:目的:研究己酮可可碱,(pentoxifylline,PTX)的抗炎作用。方法:随机将培养人脐静脉内皮细胞(EVC304)分为正常组、对照组(内毒素组)、用药组(内毒素加已酮可可碱组),各组细胞间粘附分子(ICAM-1)表达用流式细胞仪洲定,培养上清中血小板活化因子(Platelet ctive factor,PAF)用生物法测定,培养上清中一氧化氮的变化用Griese法测定。结果:内毒素加已酮可可碱组与内毒素组相比,各指标均较内毒素组有不同程度降低,但仍高于正常组。结论:已酮可可碱对炎症介质产生及粘附分子表达有抑制作用,应是其抗炎作用的机理之一。

Abstract:OBJECTIVE To investigate the effect of inhibiting inflammatory reaction of pentoxyfylline. METHODS The cell culture platelets on which the vascular endothelial cell (EVC304) grew were added with endotoxin. And some of them included pentoxyfylline. After different period of incubation, the expression of ICAM 1 on vascular endothelial cell membrane and the NO and PAF in media were assayed. RESULTS Comparing with endotoxin group, the expression of ICAM in pentoxyfylline group reduced, so did PAF and NO. CONCLUSIONS Pentoxyfylline can inhibit inflammatory of endothelial cell by reducing the expression of adhesion molecualr and the releasing of some inflammatory media.
